Spatially variant PSF modeling in confocal macroscopy

چکیده
Point spread function (PSF) plays an essential role in image reconstruction. In the context of confocal microscopy, optical performance degrades towards the edge of the field of view as astigmatism, coma and vignetting. Thus, one should expect the related artifacts to be even stronger in macroscopy, where the field of view is much larger. The field aberrations in macroscopy fluorescence imaging system was observed to be symmetrical and to increase with the distance from the center of the field of view. In this paper we propose an experiment and an optimization method for assessing the center of the field of view. The obtained results constitute a step towards reducing the number of parameters in macroscopy PSF model. Point spread function modelling, confocal imaging systems calibration, parameter estimation.

A framework for fast multiview fusion of Single Plane Illumination Microscopy (SPIM) images based on a spatially-variant point spread function (PSF) model recorded using fluorescent point markers (beads) inserted into the surrounding medium is presented. The beads are used for the registration of the images as well as for the estimation of the spatially-varying PSF function. A new fusion algori...
متن کاملA POSTPROCESSING METHOD FOR SPATIALLY VARIANT POINT SPREAD FUNCTION COMPENSATION by
In medical imaging such as Single Photon Emission Computed Tomography (SPECT), the point spread function (PSF) of the system is spatially variant. The current method of compensation is to model the PSF in the iterative reconstruction process of the image itself. However, this can be quite computationally expensive and may require a large amount of time to process on readily available computers ...
متن کاملImproved image quality in pinhole SPECT by accurate modeling of the point spread function in low magnification systems.
PURPOSE Single photon emission computed tomography (SPECT) has become an important noninvasive imaging technique in small-animal research. Due to the high resolution required in small-animal SPECT systems, the spatially variant system response needs to be included in the reconstruction algorithm. Accurate modeling of the system response should result in a major improvement in the quality of rec...
متن کاملMassively Parallel Spatially-Variant Maximum Likelihood Restoration of Hubble Space Telescope Imagery
We present results of concurrent maximum likelihood restoration implementations with spatially-variant point spread function (SV-PSF) on both synthetic and real datasets from the Hubble Space Telescope (HST). We demonstrate that SV-PSF restoration exhibits superior performance compared to restoration with a spatially invariant PSF. We realize concurrency on a network of Unix workstations, and a...
متن کاملSemi-Blind Spatially-Variant Deconvolution in Optical Microscopy with Local Point Spread Function Estimation By Use Of Convolutional Neural Networks
We present a semi-blind, spatially-variant deconvolution technique aimed at optical microscopy that combines a local estimation step of the point spread function (PSF) and deconvolution using a spatially variant, regularized Richardson-Lucy algorithm [1]. To find the local PSF map in a computationally tractable way, we train a convolutional neural network to perform regression of an optical par...
